Current Technical Approaches to Study RNA–Protein Interactions in mRNAs and Long Non-Coding RNAs

چکیده
It is commonly understood that RNA-binding proteins crucially determine the fate of their target RNAs. Vice versa, RNAs are becoming increasingly recognized for functions in protein regulation and dynamics RNA-protein complexes. Long non-coding emerging as potent regulators exert unknown properties moonlighting functions. A vast array RNA- protein-centric techniques have been developed identification RNA targets, respectively, including unbiased mass spectrometry next-generation sequencing readout. Determining true physiological targets challenging RNA–protein interaction highly dynamic, tissue- cell-type-specific, changes with environment. Here I review current analysis interactions living cells vitro. RNA-centric presented on basis cross-linking or use alternative approaches. Protein-centric approaches discussed combination high-throughput sequencing. Finally, impact mutations complexes human disease highlighted.

The Role of Long Non-Coding RNAs in Ovarian Cancer
Background: Ovarian cancer is the most fatal tumor of female's reproductive system, and several genetics and environmental factors are involved in its development. Various studies have already identified suitable biomarkers to facilitate the early detection, prognosis evaluation, and the assessment of treatment response. However, the aim of this review was to investigate the role of long non-co...
متن کاملLong non-coding RNAs and their significance in human diseases
Protein-coding genes account for only a small fraction of the human genome and most of the genomic sequences are transcriptionally silent, but recent observations indicate significant functional elements, including non-coding protein transcripts in the human genome. Long non-coding RNAs (lncRNAs) have been defined as transcripts of >200 nucleotides without protein-coding capacity that perform t...

متن کاملIdentification and Functional Prediction of Long Non-Coding RNAs Responsive to Drought stress in Lens culinaris L.
Drought stress is one of the main environmental factors that affects growth and productivity of crop plants, including lentil. In the course of evolution evolution, crucial genetic regulations mediated by non-coding RNAs (ncRNAs) have emerged in plant in response to drought and other abiotic stresses. In the present study, after identifying lncRNAs within the expression profile of lentil, RNA-s...
متن کاملExperimental approaches to identify non-coding RNAs
Cellular RNAs that do not function as messenger RNAs (mRNAs), transfer RNAs (tRNAs) or ribosomal RNAs (rRNAs) comprise a diverse class of molecules that are commonly referred to as non-protein-coding RNAs (ncRNAs). These molecules have been known for quite a while, but their importance was not fully appreciated until recent genome-wide searches discovered thousands of these molecules and their ...
